The two-vehicle collision at Frog End happened at about 12.30pm.
John Faint, 51, of Sawston, who was travelling in a Peugeot 206 and Mary Gray, 54, from Winchester, who was travelling in a Vauxhall Zafira MPV, suffered fatal injuries.
Two other people, one in each car, were treated at Addenbrooke's Hospital, Cambridge. The A10 between Royston and junction 11 of M11 was closed for five hours and traffic was diverted.
Pc Pete Bimson said "We would like to appeal to the driver of a dark vehicle who stopped and helped following the collision.
"We would also like to hear from anyone else who may have witnessed anything."
